答:每个frame的剂量太低,没法准确align,motion correction也就失效了。High frame rate means high electron dose rate. High electron dose rate means …
那么,如果成像速率足够高, 是否可以在样品损伤前,把图像信息收到?
猜想:dose可以变高的话,那么每个frame就算时间很短,也有足够的信息做alignment。这样也许就可以更充分得sample 样品的漂移,也就可能带来更精准的motion correction。
引发:XFEL是如何解决这个问题的?
答:XFEL 是样品快速流过射线源,样品全靠shutter保护。
